The Steinmetz group at EMBL Heidelberg seeks an ambitious experimental postdoctoral fellow to lead an interdisciplinary project, focusing on genome editing technologies for functional genomics. The appointed candidate will establish a precision gene editing system with the aim to chromosomally tag proteins in high throughput. The technology will be used in close collaboration with the team of Michael Knop (University of Heidelberg) to perform high-resolution imaging of the mammalian proteome.  

Your role

The project entails generation of new genetic tools for highly multiplexed protein-tagging and their application for confocal microscopy. You will lead the study design and organization across sites, as well as perform and analyze experiments. We have recently improved gene editing efficiency in S. cerevisiae by actively recruiting donor DNA to Cas9-mediated double-strand breaks (Roy, Smith, Vonesch et al., 2018). This allows us to systematically generate genetic variants in the yeast genome and measure effects in different environments.

We seek a postdoctoral candidate interested in developing genetic tools with improved homology-directed repair and precision editing in mammalian cells. This will enable endogenous tagging of protein-coding sequences - including open reading frames with unknown function and ones that cannot be identified in mass spectrometry approaches. The tool will be used in combination with high-content imaging together with the team of Michael Knop (University of Heidelberg), revealing localization, abundance, kinetics and cell-to-cell heterogeneity of the mammalian proteome.  

You have

Candidates should hold a Ph.D. in a relevant field with experience in cell culture, transfection and cloning of mammalian expression vectors. Experience with genome editing technologies (CRISPR, Talens, Zinc Fingers, etc.), DNA repair, next generation sequencing analysis and confocal imaging is highly appreciated.

The appointed candidate will be independent but also enjoy working in a friendly, collaborative lab. The post holder should be enthusiastic about building and maintaining strong collaborations with the Steinmetz lab at Stanford University and the Knop group. Ideal candidate should be creative, highly motivated and well-positioned to lead a scientific project at all stages, from experimental design to manuscript preparation.
